TMED4
TMED4，即跨膜p24運輸?shù)鞍?。別名HNLF、ERS25、p24a3。TMED4在多種細胞過程中發(fā)揮作用，包括內(nèi)質(zhì)網(wǎng)應(yīng)激反應(yīng)、熱休克反應(yīng)、細胞凋亡以及囊泡蛋白運輸，主要在早期分泌途徑中。
TMED4的作用機制涉及內(nèi)質(zhì)網(wǎng)應(yīng)激(ERS)的調(diào)節(jié)，通過IRE1α-XBP1信號通路和NRF2相關(guān)的抗氧化反應(yīng)，促進調(diào)節(jié)性T細胞(Treg)中活性氧(ROS)的穩(wěn)態(tài)，維持Treg細胞Foxp3的表達及抑制功能。TMED4對于維持Treg細胞的穩(wěn)定性和功能至關(guān)重要，其缺失會導致Treg細胞中Foxp3穩(wěn)定性降低，影響Treg細胞的抑制功能，進而可能導致T細胞過度激活和炎癥反應(yīng)加劇。TMED4還可能通過ER相關(guān)的降解(ERAD)系統(tǒng)抑制IRE1α的蛋白酶體降解。TMED4的功能異常與多種疾病相關(guān)，包括神經(jīng)細胞脂褐素沉積癥8型。
