GPR20
GPR20，全稱為G protein-coupled receptor 20，是一種在多種組織中表達的孤兒GPCR。該蛋白在胃腸道間質瘤（GIST）中高度表達，成為治療GIST的非激酶新靶點。研究表明，GPR20在GIST細胞系和患者來源異種移植（PDX）模型中表達顯著增加。基于這一發現，研究者開發了靶向GPR20的抗體藥物偶聯物（ADC）DS-6157a，該藥物在臨床前模型中表現出依賴GPR20表達的抗腫瘤活性，對TKI耐藥的GIST有效。DS-6157a的臨床前評估顯示其對TKI耐藥的GIST模型具有顯著抗腫瘤活性。此外，早期臨床試驗表明，DS-6157a的藥代動力學特性與安全性之間存在明顯的關系，藥物暴露量與中性粒細胞減少癥和血小板減少癥等血液學不良事件相關。盡管DS-6157a的研發已被取消，但第一三共擁有強大的ADC藥物管線，包括Enhertu等。
