GFRA1

GFRα1(膠質細胞源性神經營養因子受體α1)是GDNF家族配體的核心受體之一,通過與RET酪氨酸激酶受體形成復合物調控神經元存活與分化。該蛋白在胚胎發育期對交感神經、腎臟及腸道神經系統的形成起關鍵作用,其功能異常可能引發先天性巨結腸癥、多巴胺能神經元退化等疾病。近年研究發現GFRα1在甲狀腺癌、神經母細胞瘤中存在異常表達,其介導的RET信號過度激活與腫瘤侵襲轉移密切相關。針對這一通路開發的凡德他尼、卡博替尼等RET抑制劑已獲批用于甲狀腺髓樣癌治療,而基于GDNF-GFRα1系統的基因療法在帕金森病動物模型中展現出神經保護潛力。目前重組GDNF蛋白的鞘內給藥方案正在進行Ⅱ期臨床試驗,旨在為肌萎縮側索硬化等神經退行性疾病提供新治療策略。
